Identification and Cloning of a Putative Male Fertility Gene Encoding an Oxidosqualene Cyclase in Qingke

نویسندگان

چکیده

Anther development is crucial for controlling crop fertility. To elucidate the underlying mechanisms of reproductive highland barley (called Qingke in Chinese), two main cultivars, Zangqing 2000 and Ximala 22, were utilized. Transcriptome analysis showed that lipid, sugar phenylpropane metabolisms might be major pathways associated with male fertility by analyzing possible common DEGs before anther maturation both varieties. Additionally, 26 genes related to genic sterility screened identify homologous sterile lines. Among them, HORVU.MOREX.r3.7HG0634780 (HvtOSC12), an oxidosqualene cyclase, was highlighted as a candidate gene affecting fertility, it highly especially expressed maturation. Furthermore, HvtOSC12 (including promoter sequence) cloned homology-based cloning. The further bioinformatic deduced MYBs one transcription factors expression responding environmental changes. These results lay foundation potential applications creation environment-sensitive Qingke.

Cloning and molecular characterization of TaERF6, a gene encoding a bread wheat ethylene response factor

Ethylene response factor proteins are important for regulating gene expression under different stresses. Different isoforms for ERF have previously isolated from bread wheat (Triticum aestivum L.) and related genera and called from TaERF1 to TaERF5. We isolated, cloned and molecular characterized a novel one based on TdERF1, an isoform in durum wheat (Tri...

Conversion of a plant oxidosqualene-cycloartenol synthase to an oxidosqualene-lanosterol cyclase by random mutagenesis.

A random mutagenesis/in vivo selection approach was applied to generate and identify mutations that alter the product specificity of oxidosqualene-cycloartenol synthase (CAS) from Arabidopsis thaliana. This work complements previous studies of triterpene cyclase enzymes and was undertaken to provide knowledge of the frequency and locations at which point mutations can alter cyclase product spec...

Cloning and characterization of CSP37, a novel gene encoding a putative membrane protein of Candida albicans.

In the course of an analysis of the functions and assembly of the cell wall of Candida albicans, we have cloned and characterized a gene, which we designated CSP37 (cell surface protein), encoding a 37-kDa polypeptide which is a membrane-associated protein. The gene was isolated by immunological screening of a DNA library constructed from mycelial cells with a polyclonal serum raised against ce...

Cloning of the Gene Encoding M2e of Influenza Virus in B. subtilis

Background and Aims: The ectodomain of matrix protein of influenza virus is a weak immunogen that is highly conserved among all subtypes of influenza A virus. Tandem repeats of these genes along with linker were used to enhance immunogenicity of M2e protein and so it can be served as a universal vaccine in both humans and livestock. Materials and Methods: In this study, the sequences of extra-d...

Cloning and expression of a gene from Streptomyces scabies encoding a putative pathogenicity factor.

We cloned a 9.4-kb DNA fragment from Streptomyces scabies ATCC 41973 that allows the nonpathogen Streptomyces lividans 66 TK24 to necrotize and colonize potato tuber slices and produce scab-like symptoms on potato minitubers. Deletion analysis demonstrated that activity was conferred by a 1.6-kb DNA region. Sequence analysis of a 2.4-kb DNA fragment spanning the DNA region necessary for activit...
